Pullman and Tur Bus both operate out of La Serena and Coquimbo. Buses are available both south to Valparaiso and Santiago and north as far as Iquique. Buses leave every 20 minutes for Ovalle from the main terminals.
Via Elqui and Sol de Elqui leave frequently for Vicuña and other towns in the valley from the bus station. Although there’s no shortage of buses that will take you to many of the valley’s places of interest, if you have the luxury of a private vehicle, then take advantage of stops along the valley’s winding roads. The Embalse Puclaro, for instance, is an expansive glacially fed reservoir with a large wind harp that sends eerie harmonic tones echoing through the valley. You can purchase a tourist pass from La Serena’s terminal, which will allow you to get off and on buses as you please.
