El Panecillo, or to use the Inca name, Yavirac, is a lush green hill visible from many different points in Quito. La Virgen del Panecillo, the winged virgin sitting atop the hill, is essentially the main religious symbol of Quito. As a result, when visiting the monument, guides tend to quote scriptures as opposed to emphasizing historical or structural facts. The view is incredible, especially on a clear day, so even if aren't interested in listening to a recital of Revelations 12, you will enjoy the trip.
The best way to get there is by cab. From the Centro Histórico, you shouldn't pay much more than $5. Alternatively, you can arrange a tour that includes a trip to the Panecillo at any travel agency. The bus that goes up to the Panecillo is the same green bus you can take to Mitad del Mundo (but on the southern end of the loop, obviously). Be careful at the bottom of the hill and on the roads leading up the Panecillo as there have been many reports of mugging.
